Coleen: I neglected to answer the temperature question. Using paraformaldehyde at 4C is more tradition than anything. We found that there are more undesirable ultrastructural changes using cold paraformaldehyde than you see when the tissue and the solutions are at approximately the same temperature. Freida Carson
