Hello Histonet,
Morphosave is available from Ventana. It is cat. item 250-010, and
comes in a 500ml bottle of 10X solution. We use it at a 1X dilution,
which we keep for one week at a time. Slides are frozen sectioned and
immediately placed into cold acetone for 15 mins. Slides are then
allowed to dry completely, and placed in dessicant for at least an hour.
Just before staining, slides are again placed into cold acetone for 5
mins., air dried, placed into 1X morphosave for 15 mins., and rinsed
with buffer. Staining continues as usual from this point. In our
experience, DAB staining doesn't seem to be greatly enhanced, but the
morphology in Alk. Phos staining is greatly improved.
