Plate reader for substance P and CGRP

From:Clover Daley

I need to set up a procedure for measuring substance P and (CGRP) Calcitonin
Gene Related Peptide in rat spinal cord. This is what I have found in the
literature. If anyone has done this I would appreciate talking with you.

Tissue extracts are prepared by cryosection of the cord at 20 microns.
collected sections are added to 0.5M glacial acetic acid, then placed in a
boiling water bath for 10 min. After removal of particulate debris by
centrifugation, the acetic acid supernatant is lyophilized using a Speed-Vac
concentrator.

The lyophilized samples are then resuspended in the assay buffer and assay
immediately for SP and CGRP. A plate reader is then used to measure the
concentrations.
