muscle section autofluorescence

From:"Abigail.Mackey"

Hi,
I am staining for gelatinase enzyme activity in human skeletal muscle
sections, using gelatin conjugated to a fluorescent green marker (Oregon
Green Molecular Probes). There appears to be some background fluorescence
and I'm wondering if it's possible to eliminate this completely or is it
something I just have to live with? (The tissue is not fixed.) I've searched
the archives and found some information on reducing autofluorescence due to
the fixative used. Any advice for unfixed tissue?
many thanks,
Abigail
