Re: ? about Bouins fixative

From:Gayle Callis <uvsgc@montana.edu>

For an elastic stain, I would be using a different method to identify
elastic fibers specifically, Weigerts Resorcin-fuckshin our favorite for
staining finer elastic fibers, or Verhoeffs van Giesons. NBF formalin was
used for fixation, never Bouins for elastic fibers.

I think you are asking about Bouins in conjunction with Massons Trichrome
for connective tissue (includes collagen, elastic and basement membranes
which will stain in shades of blue with aniline blue or green with light
green (?). So questions will be answered in terms of Masson Trichrome.

I would like to know if there is any difference with staining results:
On paraffin sections using any one of these steps.

1. If you first fix the tissue in Bouins.
2. If you use NBF fixed cut section on a slide in Bouins in an oven for 1hr.

**** I found no differences in my Mass tri staining, and preferred to do
post NBF fixation mordant with Bouins rather than fix fresh tissues in Bouins.

3. If you use NBF fixed cut section on a slide in Bouins on the countertop
    overnight.

I prefered overnight RT Bouins mordant technic for overly large decalcified
bone sections. These sections stayed on the slide better than using heated
(oven method) Bouins postfixation.

End results remained the same for all three ways to use Bouins.

I know all three steps will work but will the end result stain better.
I am using a elestic stain.
