I have found some antigens respond well to pressure cooking and microwave, B5
fixed tissue does not, other clones like enzyme and hate steam or pressure or
microwave, I have been doing all three or should I say 4 if we include
enzymes for about 5 yrs and it is all trial and error, clone dependent,
fixation driven, dtection chemistry sensitive. So if you enjoy , discovery,
playing with antibodies, and trial and error , have fun. Please be aware that
you can over retrieve as well and force false positivity, I did it with some
liver tissue and a clone in test cases , confirmed by pcr.
This is a great field. Dana
