Dear histonetters,
I have been processing mouse carotid arteries placed in Histo screen biopsy casettes (Shandon) after fixation in 1% paraformaldehyde (+-1hr.). I process them in a Shandon Citadel 2000 for 30 min. in each bath (starting 70% etoh , 80%, 95%X2, 100% x2, Histoclear (Amresco) x2and paraffin (vacuum) x2. I am consistently getting "beef jerky" impossible to section tissue, any ideas on what I'm doing wrong? I will try any suggestions!!!
Thanks,
Alexis
