Hi Susan:
  I always used nickel salts when I wanted to detect 2 different epitopes in the same section.
  One I detected with regular DAB to get the "classical" permanent brown-reddish color. To detect the second epitope I used a DAB modification with nickel salts that will give an also permanent signal, but this time "blue-purplish" in a way that I could visualize both in the same section.
  I prepared the second chromogen as follows:
  a- DAB ---- 6 mg
  b- PBS ---- 10 mL
  c- 3% H2O2 --- 160 µL
  d- 16.2% aq. solution of nickel sulfate hexahydrated --- 120 µL
  Mix like the regular DAB and apply to the section an let act for 6 minutes.
