Re: azure B

From:rueggp

i use azure b all the time but not to stain melanin. i use it to stain bone, cartilage and collagen.
it is a very simple stain
0.2% azure b
i use it at different ph's for different tissue components
for you i would use at ph 6-7, then it stains about everything
if you just wanted cartilage to stain use an acid ph about 1.2
ph 4 will stain cartilage and bone
ph 7 stains most components

make up the stain and ph it to what you want
stain the sections for 5 min., pour off the stain and save it (do not filter)
rinse the slides til clear in deionized h20
at this point i air dry and coverslip with permount without going thru alcohols and xylene, the solvents will remove the stain
we love this stain, it is so easy and versatile
