I am posting this for a colleague who is doing immunohistochemistry on
frozen sections of mouse intestine (OCT embedded). She is having a
terrible time keeping the tissue on slides during IHC and keeping the
morphology good, especially the villi tips. She has tried a number of
different kinds of coated slides - including vectabond, poly-L-lysine and
plus slides, both with and without air drying after sectioning. She is
not doing any antigen retrieval. Any suggestions on how to get tiny pieces
of mouse tissue to stay on slides during IHC and preserving the morphology?
