We are also using the Novocastra CD10 antibody (clone 56C6, catalog#
NCL-CD10-270). We use antigen retrieval in 6mM citrate, pH 6.0, in a
microwave pressure cooker for 10 minutes at pressure. The antibody
dilution is 1:10, and we use Vector ABC Elite detection. We are
generally getting good results, but if we have a "weak" day for any
reason, this antibody is one of the first to suffer. At 1:10, there
isn't much room for error.
Hope this helps
