Here's the Oil Red O that I've used, but should be used on frozen sections
because regular processing disintegrates the fat.
Oil Red O
Fixative: None
Technique: Cut frozen sections at 4 to 6 micronsl.
Quality Control: Tissue containing fat.
Staining Procedure
1. Stain slides in working Oil Red O for 10-15 min
2. Wash in tap water.
3. Mayer's Hematoxylin for 10-30 sec.
4. Wash rinning water for 3-5 min
5. Mount with water based mounting media.
Results
Fat...........................red
Nuclei.....................blue
Solutions
Oil Red O (stock)
Oil Red O.......................0.5 gm
Isopropyl Alcohol...........100 ml
Oil Red O (working)
Oil REd O (stock)...............6.0 ml
Distilled water.....................4.0 ml
mix just before use and let stand 7-10 min, filter
