I prefer to use an alkaline phosphatase StreptABC system for melanoma
markers, using Dako's New Fuchsin substrate/chromogen kit.
This gives a vivid red stain which contrasts well with brown melanin
pigment.
As a matter of interest Dako claim their New Fuchsin chromogen is Xylene
stable - I find Xylene reduces the intensity of product (although it doesn't
remove it completely), so I mount from alcohol in Shandon Xylene-substitute
mountant which retains the bright red colour.
