I have worked with Ventanas ES and Nexes systems and have never needed
amplifier for either antibody, so it does not surprise me that you get high
background. Also Nse ( humorously referred to as Non-specific Enolase) can
just stain lots of stuff it shouldn't so I can see why you are getting high
background. Try using nothing but detection and I an sure it will work fine.
