My comments to Andi were certainly not directed as a criticism of his
reference to zinc formalin instead of the "zinc fixative". I did the
same thing with this whole zinc fixative scenario. So I decided to
go to the source of some of this confusion, PharMingen, as they have a
wonderful protocol filled catalog, not to slight their excellent products.
My conversation with PharMingen yesterday was an attempt to have them
CLARIFY this zinc fixative as non-formalin, or formalin-free zinc fixative,
plus more put more details in their catalog. Hopefully they will add
the following on fixative description to save us time/grief when we use the
ZnTris nonformalin fixation.
It does not require antigen retrieval (lovely thing to know!)
Alkaline phosphatase is the substrate used by both authors of the
two publications Pharmingen put in their catalog
At any rate the fixative is worth a try for excellent IHC on rodent tissue,
paraffin embedded and good morphology, and NO ANTIGEN RETRIEVAL!
