In-situ microtomy

From:"Patrick M. Haley " <pmhales@cybergap.net>

Fellow Netters,
I have a researcher wanting to perform in-situ on mouse feet. They are
looking at an arthritis model. The specimens will be fixed in
Methyl-Carnoy's. I have two questions:

Can these be decalcified without loosing DNA/RNA, and what is the reagent of
choice?
If they can not be decalcified, what is the best method of microtomy to
produce usable sections?

Many Thanks!!!
