problem with in situ hybr.

From:Frouwke Kuijpers

Hi everybody!
I`m working with in situ hybridization with free floating method, by using
DIG labelled probe and with the Alcaline-Phosphatase detection method. I
noticed some problem when mounting the sections, I used two type of
embeddings, Entallan with precedent dehydratation of the sections and Kaiser
without dehydratation..in both case after covering there are kind of
precipitations colocalized with the staining. with the two embeddings the
precipitations look different, but in both are present.moreover my
impression is that the precipitation is not in the section but above it. I
would like to ask if any of you had similar problems and if you have any
idea of how to solve such a problem
