Hi Catherine,
I am no expert but I have a little experience. In human tissues I had 
to use heat retreival on my formalin-fixed, paraffin-embedded 
tissues as well as leaving the antibody on overnight at 4 C. In 
canine tissues I couldn't get it to work at all (although someone else 
at this institution could, but I don't know the details), and in rats it 
worked like a charm with ag retreival not required and incubation 
for only 1 hr at room temperature! There are many different COX-2 
ab's out there and as many or even more different sources. I think 
the trick might be to find the one that suits your application best.
