Re: p16 immuno staining
Could you specify the clone? We have been using mAbs against p16alpha from
NeoMarkers (clone DCS 50.2) with little success and from Pharmingen (clone
G175-405; working conc. 5 ug Ig/ml, HIER in citrate buffer pH 6.0, pressure
cooker) with a little more success. While p16alpha is considered to be a
nuclear antigen, it seems that reactivity in the cytoplasm can occur (see
e.g. Gugger et al. 2001, Lung Cancer 33:229-39; significance?) - this is
probably what you identified as background.
