RE: C4d staining for renal biopsies

From:"Sebree Linda A."

Hi Cynthia,

We use Biogenesis C4d Ab (cat. # 2222-8004) and have worked it up for IHC.  We use it at 1:600 on Ventana instruments at 24" to 32" depending on which instrument.

Hi Martha,

I have researched the C4d Ab, and it only comes as a fluorescent Ab.  It's an indirect IF procedure, only doing it on fresh, frozen kidney bx's.  We have had good luck with the procedure so far, having 1 positive case.  You can purchase the Ab with Biogeneis.
Good luck!

Martha,

We only do C4d on frozen renal biopsies  mainly because the clinicians want their answer sooner rather than later.

Our lab has had inquiries about adding this antibody to our protocol for transplant renal biopsies and I wanted to hear from labs that are currently performing it.  All the protocols I have seen reference frozen tissue but we were wondering if there is a way to do it on ffpe tissues.  Any and all help will be greatly appreciated!
