My procedure is as follows:

TRIS HCl buffer:
TRIS HCl ....... 6.61 g
DI water ...... 1 liter
Prepare in a 1 Liter volumetric flask
 
Protease solution:
TRIS HCl ....... 100 mL
CaCl ............  0.025 g
dissolve CaCl completely, then add 0.025% Trypsin and dissolve.  

Incubate sections at room temp for 6 minutes.
