There are methods for low temperature/longer time antigen retrieval, Jules
Elias wrote a paper on this very thing for Journal of Histotechnology, 1997
20(2)155-158..  There are probably more low temp methods published since then

As for temperature, do you mean NO increase in temperature as enzyme
digestion is normally carried out at 37C, a bit above RT but not excessive
