Hello, I have questions concerning a new project I am starting. It
involves human arteries. The tissue is already frozen but not fixed when I
receive it. My job is to cut cross sections and perform basic H&E and
immunofluorescence. I have previously only worked with decalcified fixed
tissue which I embed in O.C.T. My questions are: How do I embed it or do
I need to embed in order to cut it? What temperature? How thick? Also, do
I need to fix on the slide and with what fixative? I understand that some
fixatives might inhibit my antigen detection? All the references I can
find so far refer to cultures or smears, not frozen sections on slides. I
would appreciate any help or advise.
