RE: [Histonet] melanin pigment

From:"Edmondson David \(RBV\) NHS Christie Tr"

Last week we used a Mallory bleach to remove the melanin, which worked
well enough for Mart-1(Melan A) but not for HMB45. We then use a VIP
purple DAB product ( from Vector) on another set of slides without the
bleach. Whatever it takes

I don't recall this question, but I would be interested in how you get
around the problem of epidermal pigmentation if you are looking for
protein expression in the epidermis; do you bleach the tissue (and if
so, does that affect antigenicity of the proteins), or can you choose a
detection chromogen that has a wild color, like a Barney purple color?

Thanks for your help.

Mel

We use Vectors Nova Red and it works well with melanin and also
hemosiderin.
