Sharon,
              I agree with that the sectioning of spleen perpendicular to
the long axis is a desired one. I also suggest you to refer the following
paper. This contains a standarised methodology for sectioning and trimming
of all the organs of mice and rat to nullify the variability among labs and
evaluation. Good luck.
