Re: Periodic acid methanamine silver staining question

From:Rena Fail

basement membrane in the glomeruli stains last. Usually only takes 40 minutes in a 60 degree oven.  We use a microwave to hasten the silver staining. 1200 watts 50 percent power 40 seconds. check the slides when they are a lt tan in color  check microscopically for end point,  keep the silver warm by reheating for 20 seconds in the microwave. takes from 1-4 minutes to stain.  reference in Sam Spicer's Histochemistry in Pathologic diagnosis

Hi all-

I am having a problem getting the basement membranes in the glomeruli to
stain.  The basement membranes of tubules and reticular fibers are staining
very well.  I'm using a kit from Polyscientific, Inc that recommends
incubation in at 70C for 60-75 minutes.  I've gone out to 90+ minutes with
no improvement.  I was hoping somebody out there would be able to shed some
light on the subject.  I'm going to another protocol that calls for
microwaving at 95C for 15 to 20 minutes but I wanted to see if there were
any other thoughts out there.

Thanks for the help.
