Re: Dapi
Hi Jenny,
I don't have information about that specific nuclei stain but I do use
another fluorescence stain for nuclei: bis-Benzimide (Hoechst) . I mix 1
mg of bis-benzimide in 1 mL distilled water. Then I take 4.0 micro
liters of this stock and dilute with 1 mL PBS pH 7.4. The Sigma
catalogue number for the bis-benzimide is B 2883. I apply it to my
cryostat sections for 10 minutes and rinse 3x for 5 minutes in PBS. The
fluorescence color of the nuclei is bright blue and is viewed with a UV
filter. The stock solution should be frozen (-20 C) and kept protected
from light when not in use.
If you want a picture of what to look for let me know and I will email
one directly to you.
