Frozen Bone Sections...Help!

From:Tracey Couse

Hello to Everyone,

I am not new to frozen sectioning, but I am new to cutting frozen bone
sections. Any help to prevent me from pulling out more hair would be
greatly appreciated.

The first question is, are decent frozen bone sections possible without
using the tape transfer system? I have seen that there has been some
debate about this on histonet in the past. The tissue I am trying to cut
is rabbit tibia as practice for a small upcoming human project. I have
tried a range of temperatures, but -30 to -35 degrees Celsius seems to be
the suggested temperature based on the reading I have done. I am also
using a tungsten carbide knife and Superfrost Plus slides. I was hoping
for sections in the 7-10 um range. The problems are that I am getting
shattered sections and even when I try to pick up a section, the actual
bone does not want to stick to the slide. I have not yet tried other
coated slides, mind you this would only eliminate the adherence
problem. Any suggestions?!

As always, I am exceptionally grateful for any help you may provide.
