Dear linda
No problem, I assume you want me to explain , how we fix our cytospins for
retrieval purposes? We have fixed in formalin, acetone/alcohol 50/50 or
alcohol only. This depends on the antibody that needs testing. I usaually fix

at room temp for 10-15 minutes then allow to air dry and proceed with any
set
retrieval protocol that the antibody requires. I have found all methods
eliminate extraneous blood(water contents rupture cells) formalin and
alcohols fix white and malignant cells to slide. For example since
cytokeratins are a popular request after fixation we do protease on our
instrument for 8 min and then stain, if ER/PR is requested , then formalin
fix and then microwave retrieve, etc. If I can be of any additional help ,
you can e-mail me at work,(just got it DDittus@amh.org) or contact me by
phone 215-481-2609. Feel free to post this on histonet if you like.

       Dana
