beta galactosidase protocl for frozen sections

From:Gayle Callis

Greetings all,

I just realized my BetaGal staining protocol is for whole mount tissues,
and not for frozen sections.

Can someone point me in the right direction, or if you have a protocol, I
would be very grateful. The method I have is the following from a
publication, if this is correct and comparable to yours, let me know.

Frozens, fixed with 0.5% gluteraldehyde in PBS 10 min

Rinse X 2 with PBS containing 1 mM Magnesium chloride

1mg/ml XGAL solution (5mM potassium ferrIcyanide) and 1mM MgCl2 in PBS for
4 hours at 37C, counterstain with 1% neutral red.

Trying to read the buffer here, means 5mM potassium ferrIcyanide in PBS
with 1mM MgCl2.

My whole mount staining had potassium ferrOcyanide included, so staining
sections is different OR was the latter salt left out???

Thanks,
