Here's one, it's quite easy.
Fixative: 10% neutral buffered formalin.
Technique: Cut paraffin sections at 3 to 5 microns.
Quality Control: Section containing ferric iron.
Staining Procedure:
1. Deparaffinize and hydrate sections to distilled water.
2. Place slides in potassium ferrocyanide-hydrochloric acid for 10 min.
3. Rinse well in distilled water.
4. Counterstain with nuclear fast red for 5 min.
5. Rinse in distilled water.
6. Dehydrate, clear , and mount.
Results:
Ferric Iron..............................bright blue
Nuclei.............................................red
Cytoplasm...............................light pink
Solutions:
2% Potassium Ferocyanide
Potassium Ferrocyanide...........................2.0 gm
Distilled water........................................100.0 ml
2% Hydrochloric Acid
Hydrochloric Acid.....................................2.0 ml
Distilled Water.......................................98.0 ml
Potassium Ferrocyanide-Hydrochloric Acid Solution
2% Potassium Ferrocyanide......................25.0 ml
2% Hydrochloric Acid..............................25.0 ml
Nuclear Fast Red
Aluminum Sulfate........................................5.0 gm
Distilled water..........................................100.0 ml
Nuclear Fast Red........................................0.1 gm
Dissolve aluminum sulfate in distilled water, then add nuclear fast
red and dissolve with aid of heat. Cool, filter, and add a grain of
thymol as a perservative.
