Andrea,
To prepare Giemsa stock, dissolve 0.5g powder in 33ml glycerol by
standing at 50-60C for 2 hours, add 33ml methanol(acetone free) and
allow the soln. to stand 24 hours. To stain dried blood smears, they
should be fixed in methanol for 2-3 mins then transferred to diluted
Giemsa ( try 1:20-1:50 in pH6.8 phosphate buffer) for 10-20 min.
