I have done Alcian Blue/Periodic Acid Schiff's stain on GMA sections. The trick is a 10 minute pre-treatment of the slides with a Sodium Ethoxide solution to etch away the plastic. Rinse well in running water before staining. To make a sodium ethoxide solution, saturate 100% ethanol with excess amounts of sodium hydroxide. Store at 4 degrees overnight to equilibrate. The sodium hydroxide should not all go into solution. Be very careful using this stuff and use it in a hood. The solution can be reused as long as it is stored at -20 degrees and in a light tight container. Discard if color changes to brown. My protocol is for staining is the same as for paraffin after the sodium ethoxide step. Good Luck! === Catherine "Katie" Bresee Bennett Laboratory for Experimental Pathology Department of Veterinary Pathology Michigan State University *new* e-mail: bresee98@yahoo.com _________________________________________________________ Do You Yahoo!?
