[Histonet] DAPI staining methacrylate sections

From:"matt prideaux \(RI\)"

Hi everyone,

I'm trying to find a method to examine osteocyte density in areas of new
bone formation (between two calcein labels). The sections that we have
are 200 micron thick sections of mouse tibia. We have a confocal
microscope, which gives great images of the labels but the osteocytes
are not easily seen. I have heard that DAPI would be an ideal stain for
the osteocyte nuclei, but is it possible to stain methacrylate embedded
sections with DAPI and if so does anyone have a method for this?

Thanks,

Matt
