goldfish

From:Deborah Faichney

Hello all,

An enquiry from a student here. He is trying to look at the epidermis and
lateral line of 10% NBF fixed Goldfish (approx. 4-5 cm in length). The
tissues have been decalcified, post-fixation, using a mixture of
Formaldehyde/Acetic acid for 4 days.
H&E Caudal T.S steak sections are not giving good micro-anatomy. T.S steaks
posterior to the head are better, but he is still unable to obtain good
tissue sections.
I will try to oversee him cassetting his next lot of samples. I have also
suggested that he only surface decalcify- incase the poor results are due to
over decalcification. I will also cut them myself to eliminate poor
microtomy.
Incase this doesn't work, anyone out there regularly working with Goldfish
who could point us in the right direction? ( He receives the samples already
fixed, so different fixative is not an option as they are already here!!)

many thanks

Debbie Faichney
