Reposting: Probe for ISH, whole mount vs slide
Thanks to those who responded to my query regarding ISH. I'm always interested in
receiving information and protocols from those who are more expert than I.
I really need some specific information regarding probe concentration:
We have some dig-labeled riboprobe that has been used successfully on stage 11 chick embryos for whole mount techniques, and want to use it on cryosections. What, if any, changes to the probe concentration or hybridization time/temperature, etc. do we need to make? Or should it work just as well at the same concentration and hybridization time and temperature?
