With traditional histochemistry it amounts to
histochemical detection of a high concentration
of -S=S- (cystine) in insulin. An oxidizing
agent such as acidic permanganate is followed
by a cationic dye at pH < 1. The dye sticks
firmly to the oxidation product (a cysteic acid).
There's plenty of peer-reviewed literature.
If you need some, ask.
