We have a retraction problem between the muscular fiber and the connective tissue. The specimens (5mm by 7mm) are fixed in 10 % formalin overnight, then in PBS buffer until processed as follow:
-alcohol, 70%, 80%, 95%, 95%, 100%, 100%, 100% an hour for each solution
-then 2 xylene substitute an hour each,
-4 paraffin 30 minutes each.

Does anybody has experienced and fixed this kind of artefact?
