Marylou,

We use a microwave pressure cooker for most of our antibodies that require
HIER. I find a couple of problems with this method: (1) There can be a
noticeable "microwave effect" which can damage the morphology of the tissue
especially if the process is allowed to go on for too long. This is
especially noticeable in skins. (2) One must establish a protocol and stick
to it in order to achieve consistent results.

We also use an old bench top autoclave for several of our antibodies. Our ER
method in particular seems to require the autoclave for adequate results.

I have never tried steaming.
