Hi. I use citrate buffer from Zymed, in the microwave, for antigen
retrieval. We look at p53, E1A, IL-10, IFNa transgene expression. I have
never checked the temperature of the buffer during the process, and just
boil the slides in the solution for 30 seconds, then turn it off for 2
minutes, then on again for 30s. I cycle like this about 10 times. I
thought citrate buffer boiled at 95C anyway, so to check the temp is moot.
