Most HPV antibodies work best on heat retrieved tissue. For tissues which
have been in formalin for so long, I would try doubling your retrieval time
(40 minutes in a water bath) or use a pressure cooker. Also try overnight
primary antibody incubations at 4’C, diluting the primary antibody 5 to
10-fold the dilution that you would use for a 15 to 30 minute room
temperature incubation.

I am having a frustrating time trying to duplicate HPV6 AND 11
immunostaining
on tissue fixed for 1.5 months in formalin. It did work once! Any
suggestions
would be very welcome!
We have tried microwaving, trypsin, prot. k and prot. VIII.
