B-Gal staining in bone

From:Liz Lummus

Hi,

Does anyone have any protocols for B-Gal staining of
mouse growth plate/leg bone in frozen sections? What
thickness should I cut the bone to get the best
morphological results? I usually cut 20 micron
sections for tissue, but I am assuming that is much
too thick for bone! Also, what age of mouse is best? I
have cut frozen bone samples from pups up to 7 days
old, but am wondering if it is possible to use bones
from adult mice.

Thanks in advance for the input!
