Dear Edna
There are various methods of retreival, microwave, steam, steam in the 
microwave,
pressure cooker, water bath ph buffers of 6,7,8, and 10 some citrate some EDTA
the methods vary lab to lab, clone to clone , tissue to tissue. Trial and 
error, advice from us, so shoot out those antibodies and look for our 
responses, this is the best text I know.
