I am after some advice on immunohistochemistry staining on bone sections.  I
am currently trying to stain for T cells in rat ankle joints (paraffin
sections).  I need to use
antigen retrieval for the antibody and am having problems with the bone
coming off the slides.  We have tried a number of slide coatings, with
gelatin and Dako silanised slides being the best so far.  Our best antigen
retrieval method so far has been 20 minutes in the autoclave.  The amount of
tissue staying on the slides has definitely improved, but is not perfect.
Does any one have advice on IHC on bone sections.
