I have done studies examining both PCNA levels or BrdU labeling.
Personally, as the technician who does the data acquisition, I prefer
BrdU as the quantitation is easier and less subjective. BrdU
quantitation is a yes/no answer. PCNA can be present at varying levels
in the cell population being examined. Thus, quantitation for PCNA is
more subjective as the intensity levels need to be graded.
However, I agree with a previous post that it depends on what you are
looking for. If you want to know what cells are proliferating at a
specific timepoint, BrdU injections are preferred. There are other
chemicals similar to BrdU (FrdU?) and if you wanted to do a
muli-labeling protocol, it can be done with these other DNA analogs.
