CD4 and CD8a no way on formalin fixed paraffin embedded.....
B220 works great in 10% neutral buffered formalin fixed. I pretreat with Proteinase K (0.0025%) for 15 minutes at 37 degrees C and it looks great with AEC..
Gr-1 works with Proteinase pretreatment but on the tumor samples I was working with had a lot of background I'm still working on that one. The spleen sample I did was better but didn't compare to exactly with a frozen spleen so I'm not exactly sure what to believe?
hope it helps.

Does anyone know if any of the following antibodies can successfully stain
paraffin embedded sections? I know the spec sheets say to use acetone
frozens, but I thought someone might have tried it anyway?

purified anti-mouse Ly-6G (Gr-1)
purified anti-mouse CD45R/B220
purified anti-mouse CD8a (Ly-2)
purified anti-mouse CD4 (L3T4)

And if not, does anyone know of a vendor that sells these antibodies that
do stain paraffin sections.

Thanks in advance.
Andrea
