Donna,
I have used the ALK1 antibody from DAKO with great success!
My control is a case of ALCL (Anaplastic Large cell Lymphoma) which is also
CD30 positive.

According to the references I have ALK1 is the "best" marker for ALCL...
detected in approx. 90% of cases. Malignant cells usually show labeling to a
similiar degree in both the cytoplasm and nucleus. I have 3-4 excellant
references if you need them.

In my lab no special "high intensity" detection is necessary.
Steam 20 min. citrate buffer, pH 6.0
Cool down 20 min.
Alk1 antibody 1:40 for 40 min. at RT
LSAB2 link and label 12 min.ea. at RT
DAB x2 for 3 min. ea.
Light hematoxylin
Techmate 1000

Hope this helps!

Subject: Anti-Human ALK Protein

Those that have used the DAKO Anti-Human ALK Protein would you please answer
a couple of questions.

1. What control tissue are you using?

2. What type of detection are you using?

We are using Ventana Nexes instruments. After talking to DAKO they
suggested using the High Intensity Detection. Has anyone tried this?

Thanks,
Donna Willis
