Histonetters:
Thanks for all of your comments and advice. I forgot to mention that my
undecalcified bone specimen includes a metal pin that will also need to be
sectioned. Based on your helpful suggestions I am planning to fix the
femurs in 4% paraformaldehyde then dehydrate in Cellosolve (Ethylene
Glycol) and embed in LR white with a cold curing UV accelerator. Hopefully
this work out, luckily I have some positive controls to try it on.
My only concern is whether or not it is ok to hold my tissues in 4%
paraformaldehyde until I am ready to embed. Is there a better non-alcohol
solution to hold them in for extended periods of time (2-3 weeks)? Thanks
again for all of your help!!
