We use DAKO's CD117 (c-Kit) pAb at a dilution of 1:200 for 30 minutes with EnVision+/HRP detection (also 30 minutes). Heat-induced epitope retrieval (HIER) is used for pretreatment. Try using a gastric stromal tumor for your positive control. See Am J Surg Path 23(4):377-389, 1999 (April) - "Interstitial Cells of Cajal as Precursors of Gastrointestinal Stromal Tumors". The antibody has worked very well in our laboratory. R. Cartun
