Alician blues are a family of polyvalent basic dyes with at least two and as
many as four isothiouronium groups per phthalocyanin ring. It works via salt
linkage with polyanions (heparain, DNA and hyaluronate) to give insoluble



Increasing concentrations of electrolytes in the staining solution results
in increased combination of Alcian blue molecules.


So it reacts by salt linkages and can be enhanced by increasing electrolyte
concentration (NaCl, KCl, LiBr, mgCl, etc)
